Resumo(s): | Os DIV-and-RAD-Interacting Factors (DRIFs) regulam diversos aspetos do desenvolvimento
das plantas através de um módulo antagonista que envolve outras duas proteínas DIVARICATA
(DIV) e RADIALIS (RAD). O objetivo principal deste trabalho é esclarecer se os AtDRIFs estão
envolvidos no desenvolvimento inicial de plântulas de Arabidopsis thaliana, em particular no
escuro. A análise morfológica de plantas mutantes atdrif crescidas no escuro foi realizada
através da medição do ângulo do gancho apical e comprimento do hipocótilo. Para investigar
os níveis de expressão de potenciais genes alvo dos DRIFs, uma análise de RNAseq foi
executada entre ecótipo selvagem (Col-0) e a triplo mutante (atdrif345) e foram identificados
genes diferencialmente expressos que envolvidos na regulação pela luz e na sinalização de
fitohormonas. As fitohormonas vegetais desempenham um papel importante durante o
desenvolvimento das plantas e, como tal, a complementação hormonal dos mutantes
atdrif345 foi realizada para averiguar se estes mutantes carecem ou excedem os níveis
normais de auxina, giberelinas e etileno. Além disso, a criação de um mutante atdrif345 com
o repórter DR5 :: GUS foi iniciada e será posteriormente usada para analisar a distribuição de
auxina no mutante triplo. Este trabalho forneceu novas pistas sobre o papel dos AtDRIFs no
desenvolvimento das plantas. The DIV-and-RAD-Interacting Factors (DRIFs) regulate diverse aspects of plant development through an antagonist module with DIVARICATA (DIV) and RADIALIS (RAD) proteins. The key objective of this work is to elucidate if the AtDRIFs are involved in the early stage of seedling development in Arabidopsis thaliana, especially in the dark. Morphological analysis of atdrif mutants grown in the dark included measurements of apical hook angle and hypocotyl length. To investigate the expression levels of several gene targets of DRIFs, an RNAseq analysis was performed between wild type (Col-0) and the triple mutant (atdrif345) and were identified differentially expressed genes, which are involved in light and hormone regulation. Plant hormones have an important role during seedling development, and as a result, hormonal complementation of atdrif345 mutants was performed to understand if these mutants lack or exceed the appropriate levels of auxin, gibberellins, and ethylene. Additionally, the creation of an atdrif345 mutant with a DR5::GUS reporter line was initiated and will be subsequently used to analyse the auxin distribution in the triple mutant. This work provided new insights on the role of AtDRIFs in plant development. |
