Resumo(s): | Os suplementos à base de cálcio têm sido amplamente utilizados para o aumento da firmeza dos
frutos e da sua resistência ao stresse abiótico. No entanto, estudos de metabolómica revelaram
que o cálcio modifica a composição da película do bago de uva e o perfil volátil do vinho, sugerindo
um efeito na microbiota nativa. O objetivo da presente dissertação consistiu em caracterizar o
efeito do cálcio na comunidade microbiana cultivável de bagos de uva maduros das castas Loureiro
e Vinhão da região DOC Vinhos Verdes, relacionando os resultados com as propriedades
bioquímicas e moleculares dos frutos. Foi ainda avaliado o efeito de diversos fatores de stresse
abiótico nos diferentes isolados da população microbiana, bem como o perfil de crescimento de
espécies de levedura isoladas. Os resultados mostraram que a aplicação de cálcio inibiu a
acumulação de antocianinas nos bagos maduros da casta Vinhão, consistente com a repressão
da expressão de genes codificantes da chalcona sintase (CHS3) e da dihidroflavonol redutase
(DFR), sem afetar o conteúdo em fenólicos totais e açúcares redutores. Verificou-se que
Aureobasidium pullulans e Hanseniaspora uvarum predominam na superfície de bagos de uva de
ambas as castas e que o tratamento com cálcio reduziu a abundância total da microbiota
cultivável, afetando a sua diversidade de forma dependente da casta. Ensaios de crescimento das
leveduras isoladas mostraram que A. pullulans é muito susceptível ao stresse osmótico, ao cálcio
e ao cobre, enquanto Wickerhamomyces anomalus é a mais tolerante ao etanol, apresentado um
metabolismo fermentativo com produção de etanol e de ácido acético. Estes estudos,
complementados com abordagens de sequenciação high throughput, revestem-se de uma elevada
importância científica e aplicada uma vez que abrem caminho, por exemplo, à produção de vinhos
com um terroir específico, produzidos por fermentação com estirpes nativas, ou à identificação de
leveduras com atividade antagonista sobre o crescimento de fungos filamentosos patogénicos,
permitindo a sua otimização em estratégias de biocontrolo. Calcium-based supplements have been widely used to increase fruit firmness and resistance to abiotic stress. However, metabolomics studies revealed that calcium modifies the composition of the grape berry skin and the volatile profile of wine, suggesting an effect on the native microbiota. The aim of this dissertation was to characterize the effect of calcium on the cultivable microbial community of mature grape berries of the Loureiro and Vinhão varieties from the DOC region Vinhos Verdes, relating the results to the biochemical and molecular properties of the fruits. The effect of different abiotic stress factors on different isolates from the microbial population was also evaluated, as well as the growth profile of isolated yeast species. The results showed that the application of calcium inhibited the accumulation of anthocyanins in the mature berries of the Vinhão variety, consistent with the repression of the expression of genes encoding chalcone synthase (CHS3) and dihydroflavonol 4-reductase (DFR), without affecting the content of total phenolics and reducing sugars. It was observed that Aureobasidium pullulans and Hanseniaspora uvarum predominate on the surface of grape berries of both varieties and that the calcium treatment reduced the total abundance of the cultivable microbiota, affecting its diversity in a variety-dependent manner. Growth assays of isolated yeasts showed that A. pullulans is very susceptible to osmotic stress, calcium and copper, while Wickerhamomyces anomalus is the most tolerant to ethanol, presenting a fermentative metabolism with ethanol and acetic acid production. These studies, complemented with high-throughput sequencing approaches, are of high scientific and applied relevance as they pave the way, for example, to the production of wines with a specific terroir, produced by fermentation with native strains, or the identification of yeasts with antagonistic activity on the growth of pathogenic filamentous fungi, allowing their optimization in biocontrol strategies. |
