Resumo(s): | A oliveira (Olea europaea L.) desempenha um relevante papel socioeconómico a nível mundial.
Contudo, a obtenção da azeitona e do seu derivado – o azeite – é limitada pela doença da gafa.
Esta doença incide em cerca de 30-50% dos olivais portugueses e leva à queda prematura e/ou
mumificação dos frutos, com consequente redução de produção e/ou depreciação da qualidade
do azeite. A estratégia predominante no combate à gafa é baseada na luta química, com a
aplicação de fúngicidas cúpricos. No entanto, a atual consciencialização do impacto destes
produtos, na saúde e no meio ambiente, tem evidenciado a necessidade da criação de alternativas
mais ecológicas, como sejam as abordagens de biocontrolo. Sabe-se que o microbioma da oliveira
desempenha um papel crucial na proteção do hospedeiro contra doenças. Posto isto, este trabalho
teve como principal objetivo, avaliar as potencialidades do microbioma nativo da oliveira na luta
biológica contra a doença da gafa. Assim, a capacidade inibitória de fungos Penicillium spp.
previamente isolados de folhas e ramos de oliveiras, foi avaliada contra fungos Colletotrichum spp.
Os ensaios de antagonismo foram realizados em co-cultura in vitro. Os resultados indicaram que
as espécies de Penicillium inibem o crescimento de Colletotrichum spp. devido a mecanismos de
antibiose, induzindo alterações morfológicas nas hifas dos fungos patogénicos, como sejam a sua
ramificação ou agregação de hifas. A comunidade microbiana endofitica e epifitica da azeitona foi
também avaliada por metabarcoding, recorrendo ao barcode ITS e à plataforma Illumina MiSeq.
As comunidades fúngicas foram avaliadas em azeitonas em dois estados de maturação (verdes e
semi-maduras), provenientes de diferentes cultivares de oliveira (Cobrançosa e Madural), com
diferentes regimes de produção (agricultura biológica e produção integrada). A riqueza relativa das
ordens Hypocreales e Xylariales relacionou-se positivamente com a tolerância das azeitonas à
doença da gafa, assim como a abundância de Aureobasidium spp.. Por outro lado, a riqueza
relativa da ordem Dothideales relacionou-se positivameente com a maior suscetibilidade das
azeitonas à doença. Com este trabalho, foi possível identificar potenciais géneros e espécies
bioprotetoras da oliveira para além de Penicillium spp.. Na generalidade, este trabalho contribuiu
para realçar as variações que ocorrem na comunidade fúngica epifítica e endofítica em azeitonas,
as quais poderão contribuir para potenciar ou reduzir o desenvolvimento da doença da gafa. The olive tree (Olea europaea L.) plays an important socio-economic role worldwide, but olives are frequently affected by anthracnose (caused by Colletotrichum spp.). This disease affects 30-50% of portuguese olive groves and leads to premature fall and/or mummification of fruits, with a consequent reduction in production and/or depreciation of olive oil quality. The control of this disease is very difficult and mostly relies on the use of copper-based fungicides. The current awareness of the impact of these products on health and environment has highlighted the importance of creating ecological alternatives, such as biocontrol approaches. The plant-associated microbial community has been increasingly recognized as playing an important role in plant health. The main objective of this work was to evaluate the potential of the native microbiome of the olive tree for the biocontrol of anthracnose. Thus, the inhibitory capacity of fungi Penicillium spp. previously isolated from apparently healthy olive leaves and branches, it was evaluated against Colletotrichum spp. fungi obtained from olives that had symptoms of anthracnose. The antagonism assays were performed in vitro by dual-cultural methods and indicated that the Penicillium species inhibited the growth of Colletotrichum spp. due to antibiosis mechanisms and induced morphological changes in the Colletotricum spp. hyphae. The anomalies observed consisted on the branching or aggregation of hyphae. Using a metabarcoding approach, through the sequencing (Illumina MiSeq) of ITS amplicons, fungal diversity was determined in olives from different olive cultivars (Cobrançosa and Madural), from different production systems (organic farming and integrated production) and in two maturation stages (green and semi-ripen). The relative richness of the Hypocreales and Xylariales orders was positively related to the tolerance of olives to anthracnose, as well as the abundance of Aureobasidium spp.. On the other hand, the relative richness of the order Dothideales was positively related to the greater susceptibility of olives to disease. With this work, it was possible to identify potential genera and bioprotective species of the olive tree in addition to Penicillium spp.. In general, this work contributed to highlight the variations that occur in the epiphytic and endophytic fungal community in olives, which may contribute to potentiate or reduce the development of anthracnose. |
