Resumo(s): | The study of physical and functional interactions between molecules in living systems is of the
utmost importance for molecular biology. Protein-protein interactions (PPI) shape the architecture
of Gene Regulatory Networks (GRN) and contribute to the levels of gene expression. In Antirrhinum
majus, the asymmetry of the flower depends on the PPI between three transcription factors:
RADIALIS (RAD), DIVARICATA(DIV) and DIV-RAD-Interacting-Factors (DRIF). The pairs DIV-DRIF and
RAD-DRIF interact to form a regulatory module responsible for the asymmetry of the flower in A.
majus and for the cell expansion in tomato. Many homologs of DIV, DRIF and RAD have been found
in several plant species, capable of regulate distinct developmental traits. Given the importance of
PPI for their regulatory function, our aim was to optimize the heterologous protein expression of
DIV and DRIF homologs to use in subsequent biophysical and functional studies, and to explore
the details of the interactions by investigating which residues are involved in PPI. Based on the
conformational analysis of 3D structure predictions, in biochemical properties of the conserved
domains responsible for the interaction, and in the experimental data resulted from site-directedmutagenesis,
we have proposed a model of interaction between the proteins. O estudo de interações físicas e funcionais entre moléculas em sistemas vivos é da maior importância para a biologia molecular. As Interações Proteína-Proteína (IPP) moldam a arquitetura de Redes Regulatórias Genéticas (RRG) e contribuem para os níveis de expressão génica. Em Antirrhinum majus, a assimetria da flor depende das IPP entre três fatores de transcrição: RADIALIS (RAD), DIVARICATA(DIV) e DIV-RAD-Interacting-Factors (DRIF). Os pares DIV-DRIF e RADDRIF interagem para formar um módulo regulatório responsável pela assimetria em A. majus e pela expansão da célula em tomate. Muitos homólogos de DIV, DRIF e RAD foram encontrados em várias espécies de plantas, capazes de regular diferentes traços de desenvolvimento. Dada a importância das IPP para a sua função reguladora, o nosso objetivo foi o de otimizar a expressão heteróloga de proteínas de homólogos de DIV e DRIF para usar em estudos biofísicos e funcionais subsequentes, e explorar os detalhes da interação investigando quais os resíduos envolvidos em IPP. Com base na análise conformacional da previsão de estruturas 3D, nas propriedades bioquímicas dos domínios conservados responsáveis pela interação, e em dados experimentais resultantes de mutagénese dirigida, propusemos um modelo para a interação entre as proteínas. |
