Resumo(s): | A cisplatina é um agente antineoplásico, citotóxico e eficaz, que tem como principal
alvo o DNA. É extensivamente utilizado no tratamento de vários tumores, induzindo a morte
celular por apoptose em células com capacidade proliferativa aumentada. Infelizmente, a
resistência celular ocorre frequentemente. Muitas das células tumorais contêm mutações na
proteína p53, originando defeitos na indução da apoptose e a resistência à droga. No entanto, a
apoptose induzida pela cisplatina pode também ocorrer em células deficientes em p53. Assim, a
elucidação dos mecanismos envolvidos neste tipo de morte celular permitirá o desenvolvimento
de novas estratégias para eliminar tumores que possuam defeitos na via p53. Muitos dos estudos
realizados nesta área foram concretizados em linhas celulares animais, as quais possuem grande
variabilidade genética e são de difícil manipulação. Utilizamos, portanto, um modelo
simplificado desprovido de um ortólogo de p53 – Saccharomyces cerevisiae. Este modelo é
válido para o estudo dos fatores envolvidos na morte celular induzida pela cisplatina, uma vez
que recentemente demonstramos que a morte celular causada por esta droga é um processo ativo
em leveduras, dependente da síntese de novo de proteínas e que não conduz à perda da
integridade membranar. Vários estudos têm demonstrado que o metabolismo celular poderá
estar na base da resistência à cisplatina. Portanto, o objetivo deste trabalho foi explorar o papel
do metabolismo e da via RAS/PKA na resistência à cisplatina, usando S. cerevisiae como
modelo de estudo.
Os nossos resultados indicam que a glucose, ao contrário das fontes galactose e glicerol,
aumenta a toxicidade da cisplatina induzindo maior morte celular. Por outro lado, a ausência de
fonte de carbono protege as células da levedura da morte celular induzida pela cisplatina.
Verificámos também que a sobre-expressão de Pde2p confere maior resistência celular à
cisplatina. Avaliamos ainda o envolvimento das proteínas Ras na morte celular induzida pela
cisplatina. Este estudo revelou que o metabolismo celular assim como a via RAS/PKA parecem
estar envolvidos na resistência à cisplatina. Cisplatin is an antineoplastic, cytotoxic and effective agent that has DNA as the main target. It is used extensively in the treatment of various tumors, inducing cell death by apoptosis in cells with increased proliferative capacity. Unfortunately, cellular resistance occurs often. Many tumor cells contain mutations in the p53 protein, resulting in defects in the induction of apoptosis and drug resistance. However, cisplatin-induced apoptosis can also occur in p53- deficient cells. Thus, the elucidation of the mechanisms involved in this type of cell death allows the development of new strategies to eliminate tumors with p53 defects. Many of the studies in this area have been performed in animal cell lines, which have high genetic variability and are difficult to handle. For this reason, we used a simplified model devoid of an ortholog of p53 –Saccharomyces cerevisiae. This model is valid for the study of the factors involved in cisplatin-induced cell death, since the cell death caused by this drug is an active process in yeast, dependent on de novo synthesis of proteins and does not lead to loss of membrane integrity. Several studies have suggested that cellular metabolism may be a basis for cisplatin resistance. Therefore, the aim of this study was to explore the role of metabolism and the RAS/PKA pathways in the resistance to cisplatin, using S. cerevisiae as a model. Our results indicate that glucose, in contrast with other carbon sources, such as galactose and glycerol, increases the ability of cisplatin to induce cell death. Moreover, the lack of carbon source during treatment protects yeast cells from cell death induced by cisplatin. We have also found that Pde2p overexpression increased the cell resistance to cisplatin. Further, we also assessed the involvement of Ras proteins in cisplatin-induced cell death, by assessing the effect of their depletion or overexpression, and found they play differential roles. This study therefore revealed that cell metabolism and the RAS/PKA pathway appear to be involved in resistance to cisplatin. Further studies will be necessary to uncover how to modulate this pathway to sensitize cells to this chemotherapeutic agent. |
