Resumo(s): | Ceramide is one of the most extensively studied bioactive sphingolipid
involved in many cell-stress responses, including apoptosis, and recognized as a
pro-apoptotic molecule. The connection between the ceramide pathway and
protein kinase C (PKC), a family of serine/threonine kinases with a key role in cell
proliferation and apoptosis, has already been established by several works.
However, the role of ceramide in the regulation of individual PKC isoforms is still
not well understood.
Hence, with this thesis we intended to study the effect of the ceramide
pathway on the activity of distinct PKC isoforms in cell proliferation and death.
With this goal, we used yeast cells deficient in Isc1p, a neutral sphingomyelinase
that enables the cell to produce ceramide by degradation of complex
sphingolipids, individually expressing the mammalian classical PKC- α, novel PKC -δ or atypical PKC-ζ isoforms considered major players in carcinogenesis.
In unstressed isc1Δ cells, it was observed that PKC-α and -ζ did not interfere
with the yeast cell growth. In contrast, PKC-δ inhibited the growth of isc1Δ cells, an
effect associated with a G2/M yeast cell cycle arrest. Together, these results
indicate that the ceramide pathway distinctly interfere with the activity of PKC-α, -δ and -ζ in yeast. Moreover, since PKC-δ-induced growth inhibition was not
observed in wild-type yeast strain, this suggests that the reduction of intracellular
levels of ceramide led to the activation of PKC-δ. Therefore, these results suggest
that ceramide is an inhibitor of PKC-δ in unstressed cells.
An additional goal of this work was to analyse whether ISC1 deletion
interfered with the reported pro-apoptotic activity of PKC-α, -δ and -ζ in acetic
acid-treated yeast cells. However, for the experimental conditions used to
reproduce the resistance phenotype of transformed isc1Δ cells to acetic acidinduced
apoptosis, the described pro-apoptotic effect of PKC isoforms was
abolished. Thus, no further conclusions could be draw about this part of the work.
As a whole, the results obtained in this thesis corroborate that ceramide is an
endogenous regulator of PKC-α, -δ and -ζ, in unstressed cells. A ceramida é um dos esfingolípidos bioativos mais estudados, estando envolvida em muitas respostas celulares e sendo reconhecida pela sua atividade pró-apoptótica. A conexão entre a via da ceramida e a cinase C de proteínas (PKC), uma família de cinases de serina/treonina com um papel chave na proliferação e morte celular, foi já estabelecida em diversos trabalhos. Contudo, o papel da ceramida na regulação das diversas isoformas da PKC não está ainda completamente elucidado. Nesta tese estudou-se a função da via da ceramida na regulação da atividade das isoformas da PKC na proliferação e morte celular. Com este objetivo, utilizaram-se leveduras deficientes na proteína Isc1, envolvida na produção de ceramida através da degradação de esfingolípidos complexos, a expressar isoformas da PKC com um papel relevante na carcinogénese, como a clássica PKC- α, a nova PKC-δ ou a atípica PKC-ζ de mamífero. Os resultados obtidos mostraram que a PKC-α e -ζ não interferiram com o crescimento das leveduras isc1Δ, enquanto a PKC-δ causou uma inibição do crescimento e uma paragem do ciclo celular na fase G2/M. Isto indica que a via da ceramida interfere de forma distinta com a atividade da PKC-α, -δ e -ζ na levedura. Além disso, dado que a inibição de crescimento pela PKC-δ não foi observada na estirpe selvagem, isto sugere que a redução nos níveis intracelulares de ceramida conduz a uma ativação da PKC-δ e, portanto, que a ceramida é um inibidor da PKC-δ em células não submetidas a um estímulo apoptótico. Um outro objetivo desta tese foi analisar se a deleção no gene ISC1 interferia com a referenciada atividade pró-apoptótica da PKC-α, -δ e -ζ em células tratadas com ácido acético. Contudo, nas condições experimentais usadas para reproduzir o fenótipo de resistência à apoptose em células isc1Δ transformadas, o efeito pró-apoptótico das isoformas da PKC foi anulado. Assim, não foi possível retirar conclusões acerca desta parte do trabalho. Em suma, os resultados obtidos nesta tese corroboram que a ceramida é um regulador endógeno da PKC-α, -δ e –ζ. |
